    Noise-Induced Linearisation and Delinearisation

    It is demonstrated, by means of analogue electronic simulation and theoretically, that external noise can markedly change the character of the response of a nonlinear system to a low-frequency periodic field. In general, noise of sufficient intensity {\it linearises} the response. For certain parameter ranges in particular cases, however, an increase in the noise intensity can sometime have the opposite effect and is shown to {\it delinearise} the response.     Symmetry Transition in Thin Films of Diblock Copolymer/Homopolymer Blends

    The effect of blending small weight fractions of low molecular weight majority block homopolymer on the structure of multilayer films of spherical morphology poly(styrene-b-2vinylpyridine) [PS−P2VP] has been studied. The structure of the films was characterized with grazing-incidence small-angle X-ray scattering (GISAXS) and transmission electron microscopy (TEM). In multilayer films of PS−P2VP, competition between hexagonal packing of the spherical domains preferred at the surfaces with the BCC (110) packing preferred by the internal layers leads to a transition in the packing symmetry as the number of sphere layers (n) is increased.(1) Neat PS−P2VP exhibits hexagonal close-packed (HCP) symmetry up through n = 4, but at four layers coexistence of hexagonal and face-centered orthorhombic phases is observed. At n = n* = 5 the face-centered orthorhombic structure (FCO) is the stable phase. On increasing n further, the FCO phase continuously distorts to approach the arrangement of the BCC (110) plane. We observe that blending a small weight fraction of low molecular weight PS homopolymer with PS−P2VP suppresses this transition and stabilizes the hexagonal close-packed arrangement beyond four layers. Moreover, n* increases with increasing weight fraction of incorporated homopolymer for the small weight fractions of homopolymer used in this study. Self-consistent-field theory simulations designed to mimic the experimental system corroborate that n* is expected to increase and show that the PS homopolymer segregates to the interstices of the HCP unit cell. This suggests that the homopolymer reduces the stretching of the PS block and the free energy penalty of HCP relative to BCC inner layers. This result is consistent with the hypothesis that the excessive stretching requirement in an HCP arrangement is the cause of its higher free energy as compared to the BCC lattice

    Cryo-EM structure of the protein-conducting ERAD channel Hrd1 in complex with Hrd3.

    Misfolded endoplasmic reticulum (ER) proteins are retro-translocated through the membrane into the cytosol, where they are poly-ubiquitinated, extracted from the ER membrane, and degraded by the proteasome1-4, a pathway termed ER-associated protein degradation (ERAD). Proteins with misfolded domains in the ER lumen or membrane are discarded through the ERAD-L and -M pathways, respectively. In S. cerevisiae, both pathways require the ubiquitin ligase Hrd1, a multi-spanning membrane protein with a cytosolic RING finger domain5,6. Hrd1 is the crucial membrane component for retro-translocation7,8, but whether it forms a protein-conducting channel is unclear. Here, we report a cryo-electron microscopy (cryo-EM) structure of S. cerevisiae Hrd1 in complex with its ER luminal binding partner Hrd3. Hrd1 forms a dimer within the membrane with one or two Hrd3 molecules associated at its luminal side. Each Hrd1 molecule has eight trans-membrane segments, five of which form an aqueous cavity extending from the cytosol almost to the ER lumen, while a segment of the neighboring Hrd1 molecule forms a lateral seal. The aqueous cavity and lateral gate are reminiscent of features in protein-conducting conduits that facilitate polypeptide movement in the opposite direction, that is, from the cytosol into or across membranes9-11. Our results suggest that Hrd1 forms a retro-translocation channel for the movement of misfolded polypeptides through the ER membrane

Strictures of the neovesico-urethral anastomotic site can be treated with regular self-dilation, but high-grade strictures require a surgical procedure involving incision by electrocautery or cold knife.     Effects of hydroxyapatite and PDGF concentrations on osteoblast growth in a nanohydroxyapatite-polylactic acid composite for guided tissue regeneration

    The technique of guided tissue regeneration (GTR) has evolved over recent years in an attempt to achieve periodontal tissue regeneration by the use of a barrier membrane. However, there are significant limitations in the currently available membranes and overall outcomes may be limited. A degradable composite material was investigated as a potential GTR membrane material. Polylactic acid (PLA) and nanohydroxyapatite (nHA) composite was analysed, its bioactive potential and suitability as a carrier system for growth factors were assessed. The effect of nHA concentrations and the addition of platelet derived growth factor (PDGF) on osteoblast proliferation and differentiation was investigated. The bioactivity was dependent on the nHA concentration in the films, with more apatite deposited on films containing higher nHA content. Osteoblasts proliferated well on samples containing low nHA content and differentiated on films with higher nHA content. The composite films were able to deliver PDGF and cell proliferation increased on samples that were pre absorbed with the growth factor. nHA–PLA composite films are able to deliver active PDGF. In addition the bioactivity and cell differentiation was higher on films containing more nHA. The use of a nHA–PLA composite material containing a high concentration of nHA may be a useful material for GTR membrane as it will not only act as a barrier, but may also be able to enhance bone regeneration by delivery of biologically active molecules

    A pilot Internet "Value of Health" Panel: recruitment, participation and compliance

    Objectives To pilot using a panel of members of the public to provide preference data via the Internet Methods A stratified random sample of members of the general public was recruited and familiarised with the standard gamble procedure using an Internet based tool. Health states were perdiodically presented in "sets" corresponding to different conditions, during the study. The following were described: Recruitment (proportion of people approached who were trained); Participation (a) the proportion of people trained who provided any preferences and (b) the proportion of panel members who contributed to each "set" of values; and Compliance (the proportion, per participant, of preference tasks which were completed). The influence of covariates on these outcomes was investigated using univariate and multivariate analyses. Results A panel of 112 people was recruited. 23% of those approached (n = 5,320) responded to the invitation, and 24% of respondents (n = 1,215) were willing to participate (net = 5.5%). However, eventual recruitment rates, following training, were low (2.1% of those approached). Recruitment from areas of high socioeconomic deprivation and among ethnic minority communities was low. Eighteen sets of health state descriptions were considered over 14 months. 74% of panel members carried out at least one valuation task. People from areas of higher socioeconomic deprivation and unmarried people were less likely to participate. An average of 41% of panel members expressed preferences on each set of descriptions. Compliance ranged from 3% to 100%. Conclusion It is feasible to establish a panel of members of the general public to express preferences on a wide range of health state descriptions using the Internet, although differential recruitment and attrition are important challenges. Particular attention to recruitment and retention in areas of high socioeconomic deprivation and among ethnic minority communities is necessary. Nevertheless, the panel approach to preference measurement using the Internet offers the potential to provide specific utility data in a responsive manner for use in economic evaluations and to address some of the outstanding methodological uncertainties in this field

    A Standardised Procedure for Evaluating Creative Systems: Computational Creativity Evaluation Based on What it is to be Creative

    Computational creativity is a flourishing research area, with a variety of creative systems being produced and developed. Creativity evaluation has not kept pace with system development with an evident lack of systematic evaluation of the creativity of these systems in the literature. This is partially due to difficulties in defining what it means for a computer to be creative; indeed, there is no consensus on this for human creativity, let alone its computational equivalent. This paper proposes a Standardised Procedure for Evaluating Creative Systems (SPECS). SPECS is a three-step process: stating what it means for a particular computational system to be creative, deriving and performing tests based on these statements. To assist this process, the paper offers a collection of key components of creativity, identified empirically from discussions of human and computational creativity. Using this approach, the SPECS methodology is demonstrated through a comparative case study evaluating computational creativity systems that improvise music
